- From Massachusetts Turnpike:
Take the Massachusetts Turnpike to exit "11A" Route 495. Take Route 495 South one exit "21B" the "West Main Street Hopkinton/Upton" exit. This will put you onto West Main Street in Hopkinton. Continue on West Main Street which will turn into Hopkinton Road in Upton shortly after passing School Street in Hopkinton at the water tower on your left. Continue on Hopkinton Road, take a left onto School Street (in Upton) just after Pratt Pond on your left side. The police headquarters is located at #30 on your right side.
- From Route 495:

- From Route 140:
Take Route 140 North through Milford or Route 140 South through Grafton. Continue on Route 140 until you see the Upton Town Hall in Upton Center. Turn onto Warren Street, which is immediately to the right of the town hall. Drive up Warren Street, the police headquarters is located at the top of the hill at the corner of Warren Street and School Street.
